In 2019, PacBio introduced HiFi reads, based on a circular consensus (CCS) technique87. During SRMT sequencing the 2 strands are linked together by bell adapters to form a circular DNA template (c.f. Section 1.2.3), the central idea of CCS is to sequence this molecule multiple times by going over the circle more than once. In the resulting long sequence the known bell adapter sequences can be removed, and a consensus sequence can be built from the multiple passes over the same DNA template. This results in long-read accuracies of 99.8% to 99.9%56,87. This works because PacBio sequencing errors are mostly randomly distributed along the sequenced template (more on that in Section 1.4.2). Therefore it is unlikely that the same error will appear in multiple passes over the same template portion.
